MEDICINA DELLA COMPLESSITA'
  "NETWORK PHARMACOLOGY: new models for developing drugs"  
     
 
 
     
  Looking for the molecular determinant of a certain disease (or, in any case, of the weak spot through which countering bodily malfunction) has been the dominant paradigm in pharmacology for about a hundred years.
The drug was able, by selectively interfering with such determinant, to cure the disease and at the same time to keep side effects to a minimum. This strategy worked very well for many years, but around the 1980s it suddenly failed. There was a seeming paradox: basic knowledge in biology boomed, but marketing new drugs plunged. A quite effective hypothesis seems to be the paradigm of 'network pharmacology': the working mechanism is no longer conceived of as a linear chain, it is rather a simultaneous action on several spots of a regulating system. It is supported both through most recent outcomes of systems biology and the experience of success drugs, which are hardly explained by classical receptor mechanisms.

All these topics will be dealt with at the Istituto Superiore di Sanità on 20th November: the meeting will be attended by expert pharmacologists as well as by physicians and engineers who have thoroughly researched the relationships between topology and dynamic behaviour of reticular systems.
